PP02 Experimental characterization and mathematical modeling of lipolysis and its regulation

Triglyceride (TG) degradation involves three consecutive hydrolytic reactions catalyzed by at least three cytosolic enzymes and, possibly, a lysosomal acid lipase. A detailed understanding of the lipolytic process is a prerequisite to elucidate its role in pathogenesis and to develop potential therapeutic interventions. Despite literally thousands of studies on the physiological role of lipolysis and its complex regulation, a quantitative description of the intricate reaction processes of TG hydrolysis is missing. PP02 combines experimental and mathematical modeling expertise with a focus on lipolysis and fatty acid homeostasis to develop a comprehensive, experimentally validated mathematical model of lipolysis. This model will lead to novel quantitative insights into the hierarchy of lipases and enzymatic reactions that control TG catabolism.
